Optical Pharmacokinetics and Elastic Scattering Spectroscopy

Elastic Scattering Spectroscopy (ESS) and Optical Pharmacokinetics (OP) are both techniques which interrogate tissue with white light, delivered by point contact via optic fibre. Different source-detector separations produce probes which are optimized for different properties; ESSís small (<350microns) separation is most sensitive to scattering, making it useful for detecting morphological changes (including those associated with cancer and pre-cancer), while the OP fibresí larger separation (~2mm) minimizes the influence of scattering on path length, suiting it more to the measurement of chromophore concentrations. Here we present clinical data on scanned ESS in the detection of lymph node metastases. In breast cancer, this diagnosis, especially when performed intraoperatively, is decisive in surgical planning. In OP, we measure photosensitizer (AlS2PC) concentrations in a preclinical model. Our results suggest that spectral information could be used to elucidate drug compartmentalization, and some rather unexpected (and unexplained) effects surface.
